Quantitative Immunological Comparison of Bird Lysozymes*

چکیده

Rabbit antisera were prepared against crystalline chicken lysozyme and characterized by immunodiffusion, immunoelectrophoresis, inhibition of enzyme activity, and quantitative micro-complement fixation. Lysozyme was purified by Sephadex G-75 chromatography from the egg whites of 17 species of birds in the order Galliformes and was tested for reactivity with anti-chicken lysozyme. The chicken and its closest relative, the jungle fowl, have lysozymes that appear to be indistinguishable from each other. The lysozymes of all the other species tested (e.g. partridges, quails, and pheasants) could be distinguished from the chicken enzyme by quantitative micro-complement tiation. Immunodiffusion failed to detect most of these differences in antigenic structure. A particularly close relationship was observed between the lysozymes of the chicken, partridges, and American quails. This suggests that these lysozymes may show very small sequence differences. This finding may also have taxonomic significance.

Quantitative immunological and electrophoretic comparison of primate lysozymes.

Rabbit antisera were produced against purified human and baboon lysozymes and tested by quantitative microcomplement hation for cross-reactivity with the lysozymes present in the milks of 19 primate species. No antigenic difference was found between the lysozymes of man and chimpanzee and almost none between those of man and orangutan. Major antigenic differences were detected between human lys...

The dependence of immunological cross-reactivity upon sequence resemblance among lysozymes. II. Comparison of precipitin and micro-complement fixation results.

Rabbit antisera were prepared against seven different lysozymes purified from the egg whites of chicken, bobwhite quail, turkey, Japanese quail, ring-necked pheasant, and duck. Cross-reactivity tests by means of quantitative micro-complement fixation were conducted among all possible pairs of these lysozymes as well as with three other lysozymes. The amino acid sequences are known for five of t...

The amino acid sequence of lysozyme from kalij pheasant (Lophura leucomelana) egg-white.

The amino acid sequence of kalij pheasant lysozyme has been analyzed. From the comparison of the tryptic peptide pattern of kalij pheasant lysozyme and maps from other bird lysozymes followed by the sequencing of tryptic peptides, the amino acid sequence of kalij pheasant was found to be: KVYGRCELAAAMKRLGLDNYRGYSLGNWVCAAKYESNFNTHATNRNTDGSTDYGIL- QINSRWWCNDGKTPGSRNLCHIPCSALLSSDITASVNCAKKIVSDGNGM...

Multiple lysozymes of duck egg white.

Lysozyme was purified from a pooled sample of Peking duck egg whites by ion exchange chromatography and gel filtration. Three different lysozymes were found, and each was shown to be pure by several criteria. The three enzymes were like chicken lysozyme in molecular size and specific activity, although they could be distinguished from it, as well as from one another, by electrophoretic and immu...

An improved 96-well turbidity assay for T4 lysozyme activity

T4 lysozyme (T4L) is an important model system for investigating the relationship between protein structure and function. Despite being extensively studied, a reliable, quantitative activity assay for T4L has not been developed. Here, we present an improved T4L turbidity assay as well as an affinity-based T4L expression and purification protocol. This assay is designed for 96-well format and ut...
